Obesity is a major global health challenge and is particularly prevalent in Scotland, where associated conditions such as metabolic dysfunction‑associated fatty liver disease (MAFLD) and heart failure with preserved ejection fraction (HFpEF) contribute substantially to morbidity and mortality. Disruptions in how the liver and heart take up, process and store lipids are key contributors to these diseases, driving the metabolic and functional changes that lead to their progression.
This project aims to investigate the role of a novel regulator of lipid homeostasis using human liver (hepatocyte) and heart (cardiomyocyte) cell lines engineered through CRISPR‑mediated gene knockout and overexpression. In hepatocytes, fatty acid‑induced lipid accumulation and the expression of genes involved in lipid uptake and lipogenesis will be assessed to determine how this regulator contributes to the metabolic disturbances associated with MAFLD. In cardiomyocytes, the effects of lipotoxicity on metabolic remodelling, mitochondrial oxidative stress and antioxidant responses will be examined to define its role in mechanisms underlying obesity-associated heart failure. Overall, this project will characterise the cell‑specific actions of an emerging regulator of lipid homeostasis and evaluate the therapeutic potential, as well as possible risks, of modulating its activity.
The successful candidate will receive comprehensive training in mammalian cell culture, CRISPR‑based gene modulation, molecular biology techniques (RT‑qPCR and Western blotting), lipid quantification and live‑cell imaging. They will also become an active member of the research team and participate in joint laboratory meetings. In addition, the student will benefit from the wider activities of the Aberdeen Cardiovascular and Diabetes Centre (ACDC), which brings together academics and clinicians to support research aimed at improving the prevention and treatment of cardiometabolic disease.
