We are looking for a PhD student to investigate how mutations in the gene PHF3 disrupt human brain development. PHF3 plays a key role during neuronal differentiation by ensuring timely gene expression. PHF3 mutations have been linked with neurological defects including microcephaly and autism spectrum disorder but it remains unclear how these mutations give rise to such severe neurodevelopmental defects. In this project we want to characterize (i) the molecular mechanism of how PHF3 patient mutations impair gene expression, and (ii) neuronal differentiation phenotypes caused by PHF3 patient mutations.
The project will use stem cell and cortical organoid models combined with functional genomics approaches (TT-seq, CUT&RUN, scRNA-seq), proteomics and neuronal functionality assays (patch-clamp and calcium imaging) to uncover how disease-associated PHF3 mutations alter gene regulation during the formation of neurons. Preference will be given to candidates with experience in stem cell and organoid models of neuronal differentiation. Excellent organizational and communication skills are very important, as well as a good command of the English language. We are looking for a team player with willingness to collaborate.
The position is for 3 years with the possibility of a 1-year extension. The position is available from October 1st 2026. The salary is 2,832.10 € (brutto)/month (14x).
