Johannes Fruehauf

Johannes Fruehauf, MD, PhD, is a physician-scientist and biotech entrepreneur whose work intersects molecular biology and innovative lab infrastructure, featured in MIT Professional Education faculty profiles. He earned his MD from Goethe University Frankfurt and his PhD from the University of Heidelberg, Germany. In 2002, Fruehauf received a Junior Faculty development scholarship from the University of Heidelberg to conduct postdoctoral research at Harvard Medical School, where he advanced his expertise in RNA interference (RNAi) technologies.

Fruehauf's research specializations include RNAi therapeutics, bacterial-mediated gene silencing, and cancer biology. He has authored over 20 peer-reviewed publications and is named inventor on nine patents related to RNAi and bacterial therapeutics. Notable publications encompass 'Short hairpin RNA–expressing bacteria elicit RNA interference in mammals' (Nature Biotechnology, 2006), 'Synthesis, cellular evaluation, and mechanism of action of piperlongumine analogs in prostate cancer cells' (PNAS, 2012), 'GF-15, a Novel Inhibitor of Centrosomal Clustering, Suppresses Tumor Cell Growth' (Cancer Research, 2012), and contributions to siRNA design and avian influenza virus inhibition. His career history features key roles in biotechnology startups: Head of R&D at Cequent Pharmaceuticals, where he invented transkingdom RNAi (tkRNAi) technology and led IND filings; co-founder of ViThera Pharmaceuticals focused on engineered probiotics; founder of Cambridge BioLabs for concept validation; and co-founder of LabCentral in 2013, a shared lab facility in Kendall Square that has incubated over 500 biotech startups, fostering Cambridge's biotech ecosystem.
