MSc by Research: Study of Hydrocephalus and water channelopathy in C. elegans model
About the Project
Hydrocephalus or ‘water on the brain’ is an understudied, life-threatening condition characterised by excessive cerebrospinal fluid (CSF) buildup in the brain [1]. CSF accumulation leads to enlarged brain in children, and severe debilitating condition in adults including chronic headaches, motor function decline (difficulty eating, walking, incontinence) and cognitive decline such as visual impairment and loss of ability of smell (hyposmia) [2]. Currently, no effective medication is available for hydrocephalus, Mutations in Aquaporin (AQP) water channels, AQP-4 (Uniprot P55087) can (i) increase or (ii) decrease water permeability or (iii) reduce its ability to form tetramer in the plasma membrane [3]. Understanding the effect of *aqp-4* mutations are needed to identify targets for improving water permeability defects. We have recently established a *C. elegans* model of aquaporin dysfunction by mutating AQP-12 in worm’s nervous system and observed olfactory impairment. The M Res student will expand the model and study how 6 different mutations (present in people with hydrocephalus) cause neuronal pathology and cognitive decline in worms.
The student will acquire in-depth training in *C. elegans* maintenance, molecular biology including genetic manipulation by CRISPR CAS9, microscopy-based imaging of olfactory neurons, chemotaxis assays, and gene expression analysis by qRT-PCR [4]. The student will be provided mentoring support as well as training in scientific writing, statistics and data management in a supportive, inclusive environment.
