Offer Description
Colorectal carcinoma (CRC) frequently harbors mutations in tumor suppressor genes—including TP53 —that are associated with tumor progression and treatment resistance. Mutant p53 (mutp53) has been shown to sustain HDAC6 expression, thereby promoting DNA damage repair and resistance to endoplasmic reticulum (ER) stress via ATF6. Preliminary data suggest that ATF6 increases TRAPPC4 expression, promoting the trafficking of PD-L1 to the cell membrane. Furthermore, HDAC6 deacetylates MSH2 and promotes…
its degradation, thereby impairing mismatch repair and contributing to mutation accumulation, neoantigen production, and immune checkpoint expression. This project aims to characterize the epigenetic alterations induced by mutp53 and their contribution to therapeutic resistance and tumor immunogenicity.
