inflammatory joint diseases, including Juvenile Idiopathic Arthritis, Rheumatoid Arthritis, Osteoarthritis, Psoriatic Arthritis, and therapy-induced inflammatory arthritis.
This project will combine synovial tissue biopsies and peripheral blood samples from patients with state-of-the-art single-cell and spatial multi-omics approaches to define the cellular and molecular pathways underlying CHIK-induced joint inflammation. Using single-cell RNA sequencing, spatial transcriptomics and proteomics, and serum protein profiling, we will apply a systems biology framework to characterise the immune and stromal cell networks that drive disease.
The study will investigate the T-cell and B-cell receptor repertoires of infiltrating adaptive immune cells alongside the transcriptional programmes of innate immune populations, including monocytes, macrophages, and neutrophils. These analyses will be integrated with detailed characterisation of the synovial tissue microenvironment, including fibroblasts, endothelial cells, and extracellular matrix-associated pathways, to identify the cellular interactions that sustain inflammation and tissue damage.
The long-term goal of this research is to establish a mechanistic framework for CHIK-induced polyarthritis that will inform the development of targeted therapies. By comparing CHIK arthritis with a spectrum of immune-mediated joint diseases, this work will also provide broader insights into the shared and disease-specific mechanisms that drive chronic inflammatory arthropathies in humans.
