Offer Description
Pediatric Acute Lymphoblastic Leukemia is the most common childhood cancer (25–30% of the total) and B-cell Lymphoblastic Lymphoma is its related solid form. Both are treated with asparaginase, effective but associated with toxicities (allergies, pancreatitis) that may cause treatment discontinuation and worsen prognosis. Evidence indicates these effects might depend on the biological response to the drug. Asparaginase is degraded by proteases, particularly AEP and cathepsin B, often overexpressed in aggressive tumors. In high-risk cases, early loss of activity suggests increased degradation. We hypothesize that elevated proteases drive both toxicity and inefficacy. The project will: analyze AEP/CTSB in sera; characterize resulting peptides; test cytotoxicity on murine pancreas; develop strategies to reduce toxicity and restore efficacy.
