Driving license category B Specific Requirements Description of tasks: Research hypothesis: During a severe course of caprine arthritis encephalitis (CAE), caused by small ruminant lentivirus (SRLV), both the systemic and local mammary gland responses are stronger than during a mild course of the disease. Gene expression in goats with a severe disease course (wasting syndrome) differs from that in healthy tissues or tissues from goats with a mild course of CAE. These differences in gene expression (mRNA, protein) result from the activity of miRNA and long non-coding RNA (lncRNA), which are modulated by SRLV. Furthermore, the viral load varies between goats with mild and severe courses of CAE. The severity of the disease is dependent on the viral genotype. Finally, SRLV influences gene methylation. Research aims: To analyze genomic, transcriptomic, proteomic, and epigenomic phenomena in blood leukocytes/serum and mammary secretory tissue of SRLV-infected goats with an acute versus mild course of CAE. This study serves as a continuation and expansion of previously conducted analyses that utilized exclusively asymptomatic animals in comparison to virus-free controls. The doctoral student will carry out a doctoral project under the supervision of prof. Emilia Bagnicka as part of the NCN OPUS30 project.
