Principle investigator Dr inż. Jakub Bogusławski
Title of the project: Two-photon fluorescence lifetime ophthalmoscopy: new perspectives in the diagnosis of eye diseases (Dwufotonowa oftalmoskopia czasu życia fluorescencji: nowe perspektywy w diagnostyce chorób oczu), FENG.02.02-IP.05-0121/25.
Description of the project: The project focuses on advanced biophotonics and optoelectronics, with particular emphasis on techniques used for optical imaging of the human retina. It centers on two-photon fluorescence ophthalmoscopy employing femtosecond laser sources. This technique offers new opportunities for early diagnosis of retinal diseases and for testing innovative gene therapies.
Within the project, we will develop new technologies to enhance the efficiency of excitation and detection of two-photon fluorescence from natural retinal chromophores. The aim is to enable precise fluorescence lifetime measurements and to establish the foundation for functional retinal imaging based on the dynamics of molecular processes [Progress in Retinal and Eye Research 93, 101170 (2023); The Journal of Clinical Investigation 132 (2), e154218 (2022)].
The research and development tasks include the design and optimization of femtosecond fiber lasers tailored to the absorption spectra of key chromophores, the development of novel methods for photon counting, correlation, and fluorescence photon analysis, and the creation of solutions that increase the sensitivity, speed, and stability of in vivo measurements.
The technologies developed within the project have direct biomedical applications and address pressing challenges in retinal disease diagnostics. The research is carried out in close collaboration with the International Centre for Translational Eye Research (ICTER) in Warsaw (www.icter.pl) and the University Hospital in Jena, providing excellent laboratory facilities and a highly interdisciplinary research environment.
